The herb Cat's Claw has been determined to activate PXR, but the specific PXR-activating compounds in Cat's Claw are not currently known. With a genetically modified PXR cell line, we ascertained that the extracts from both Gou Teng and Cat's Claw induced a dose-dependent activation of PXR, resulting in the induction of CYP3A4 expression. To determine the chemical constituents of Gou Teng and Cat's Claw extracts, a metabolomic approach was subsequently applied, and then we screened for the presence of PXR activators. Extracts of both Gou Teng and Cat's Claw demonstrated the activation of PXR by four compounds: isocorynoxeine, rhynchophylline, isorhynchophylline, and corynoxeine. Moreover, isopteropodine, pteropodine, and mitraphylline were discovered as further PXR activators from the extracts of Cat's Claw. PXR activation by the seven compounds displayed half-maximal effective concentrations below 10 micromolar. Ultimately, our research identified Gou Teng as a potent PXR activator, further revealing novel PXR activators found in both Gou Teng and Cat's Claw. Practical guidance for safe usage of Gou Teng and Cat's Claw, avoiding PXR-induced herb-drug interactions, is supported by our data.
